At IMGM, we offer qPCR analysis on the Life Technologies ViiA7 Real-Time PCR System using TaqMan or SYBR Green chemistry.  We work with 96- as well as 384-well blocks and use either general or pre-spotted microfluidic plates in different formats. ddPCR – Bio-Rad QX200 Droplet Digital PCR

In IMGM’s digital PCR workflow, the reaction is partitioned in approximately 20’000 separate droplets. Thereby, each droplet contains exactly one (positive droplets) or no (negative droplets) target sequence copy. These molecules are then amplified up to 1 million-fold by TaqMan PCR generating fluorescence signals in the positive droplets only.

These signals are used to determine the ratio of positive and negative droplets, which allows calculating back to the absolute DNA amount in the initial sample. No reference sample or standard curve is required for absolute quantification of the target DNA.

Resulting digital PCR applications with increased accuracy compared to conventional PCR are e.g. detection of single nucleotide mutations, rare alleles or copy number variation as well as absolute quantification of DNA or gene expression. Convenient analysis is made possible by a huge number of predesigned PrimePCR ddPCR Assays available from Bio-Rad for common targets. Even difficult samples like cell free DNA or FFPE material are accessible as the droplet approach reduces inhibitory effects.
